Large scale identification of HLA class I peptides from human cancer cells

Abstract

Identification of HLA class I peptides from human cancer cells was carried out using a latest bioinformatics tool, which was developed for clustering the chromatography and mass spectrometry data. The peptides were separated from the HLA large subunits and analyzed by capillary RP-HPLC combined with ESI ion-trap mass spectrometry. Patterns of peptides presented by same HLA haplotype in different cell lines were compared and peptides unique to one type of cancer cells were identified. It is stated that the patterns of the HLA peptides correlate with the pattern of proteins degradation in the cells indicating the synthesis and the possible rapid degradation of the proteins.
